参附注射液联合血必净对绵羊多脏器功能障碍综合征 PCT 和 HLA-DR 水平的影响

Effect of Shenfu injection combined with Xuebijing on the levels of PCT and HLA-DR in sheep with multiple organ dysfunction syndrome

摘要: 目的 探讨参附注射液联合血必净(XBJ)对高海拔地区内毒素致绵羊多脏器功能障碍综合征( MODS) 、血浆清降钙素原( PCT)和 MHC-II 类分子 HLA-DR 水平的影响。 方法 将 24 只绵羊随机分为正常组、模型组、参附注射液组(观察 1 组)及参附注射液联合血必净组(观察 2 组) ,每组 6 只。 除正常组外,其余各组采用静脉泵入脂多糖 LPS(6 μg / kg,30 min)建立 MODS 动物模型,观察 1 组给予参附注射液静脉泵入(100 mL,20 mL / h) ,观察 2 组静脉泵入参附注射液(100 mL,20 mL / h)和血必净(Xuebijing,XBJ) (2 mL / kg) 。 观察 1 组和观察 2 组皆以 1 周为 1 疗程,共计 1 疗程。 建模后及干预后 0. 5、1、2、3、7 d,静脉采血检测 PCT、HLA-DR。 结果 与正常组比较,模型组血浆PCT 水平降低,而 HLA-DR 升高,差异有统计学意义( P< 0. 05) ;参附注射液、参附注射液+XBJ 组绵羊的血浆 PCT水平明显降低(P<0. 05) ,且观察 2 组下降最为显著( P<0. 05) ;观察 1 组、观察 2 组血浆 HLA-DR 较模型组均有所升高(P<0. 05) ,且观察 2 组升高最为明显(P<0. 05) 。 结论 参附注射液联合血必净( XBJ) 对高海拔地区内毒素致绵羊多脏器功能障碍综合征(MODS) 动物体征的改善,对肺、肝和肾等多脏器损伤的防治作用可能与降低血浆PCT,升高血浆 HLA-DR 水平有关。

Abstract: Abstract:Objective To investigate the effects of Shenfu injection combined with Xuebijing ( XBJ) on endotoxin-induced multiple organ dysfunction syndrome ( MODS ) , plasma procalcitonin ( PCT ) and MHC-II molecule HLA-DR in high altitude areas Level of influence. Method 24 sheep were randomly divided into normal group, model group, Shenfu injection group ( observation 1 group ) and Shenfu injection combined with Xuebijing group ( observation 2) . Three sheep were taken as the blank control group. Group of 6 only. Except for the normal group, the other groups used intravenous pumping of lipopolysaccharide LPS (6 μg / kg, 30 min) to establish a MODS animal model. Observation group 1 was given intravenous pumping of Shenfu injection ( 100 mL, 20 mL / h ) , Shenfu injection ( 100 mL, 20 mL / h) and Xuebijing ( XBJ ) ( 2 mL / kg ) were injected intravenously in observation group 2. Observation group 1 and observation group 2 took 1 week as a course of treatment, a total of 1 course of treatment. After model building and 0. 5, 1, 2, 3, 7 d after intervention, venous blood was collected to detect PCT and HLA-DR. Result Compared with the normal group, the plasma PCT level of the model group decreased, while the HLA-DR increased, the difference was statistically significant ( P<0. 05) ; the plasma PCT level of sheep in the Shenfu injection and Shenfu injection + XBJ group was significantly reduced ( P<0. 05) , and the decrease in the observation group 2 was the most significant ( P< 0. 05 ) ; the plasma HLA-DR of the observation group 1 and the observation group 2 were both higher than the model group ( P< 0. 05) , And the observation of the two groups was the most obvious ( P < 0. 05 ) . Conclusion Shenfu injection combined with Xuebijing ( XBJ) can improve the animal signs of multiple organ dysfunction syndrome ( MODS) caused by endotoxin in high altitude area, and prevent and treat multiple organ damages such as lung, liver and kidney. It may be related to the reduction of plasma PCT and the increase of plasma HLA-DR levels.
